Research Interest : Cell electrophysiology on the Fluid Force Microscope (FluidFM)

Scanning ion conductance microscopy (SICM) and patch-clamp are two powerful techniques allowing to study the cell electrical properties by relying on a similar electrical setup able to measure picoampere ionic currents flowing through a tip aperture. While SICM allows to image soft structures such as cells in near-physiological conditions without mechanical contact external page(Hansma et al., 1989), the patch-clamp (external pageNeher and Sakmann, 1976) has become the gold standard for measuring the activity of ion channels, which are transmembrane pore-forming proteins involved in the cell electrical activity by a controlled flow of ions through those pores. 

By combining the SICM and patch-clamp on the FluidFM as depicted underneath, the two techniques get their possibilities expanded thanks to the additional force feedback of the FluidFM, especially powerful for the gentleness of the patching or the scanning. With such an additional force control, we were able to patch beating cardiac cells (external pageOssola et al., 2015) whereas the SICM could benefit from its combination with an AFM to reduce tip/sample interactions (external pageDorwling-Carter et al., 2015).
